It really is a testament to the writer's ability to have 'can defeat anyone in a single punch' as the running joke of the manga and still make it work, only ever needing addendums of 'but first he needs to get in that punch' or 'so long as he's not holding back'. The storytelling is good because there's a tension buildup in the story as a whole, but only because we start to see the point-of-view of other characters - we see mostly Saitama early on and he's just walking around casually one-punching things to death. But yes, certainly key is the fact that whilst Saitama is an unstoppable champion of Charles Atlas, the rest of the cast are not, and the fact Saitama exists allows for the narrative to go as high as it wants for villains - and subsequently, leave the side characters struggling even harder to keep pace. The fight with the Sea King is probably the most on the dot about this, since knowing Saitama may show up eventually means we don't have to worry on 'the world' staying safe - but the characters who have to hold said Sea King off till Saitama arrives? That's a different matter entirely.

Those aren't the addendums, or even terribly important. There's not any question as to the winner of any fight Saitama's in, and pretending that he might somehow get killed by Fubuki's Hell Wind -> Psychic Tornado -> Concrete Beartrap -> Boxcutter combo string if he doesn't get serious fast enough is just silly. Onepunch Man works the same way a good Superman story does--it's not a question of whether or not he can defeat Asura Rhino, or Braniac, or Goku. It's just a story about someone who can walk into any bank vault on the planet and take absolutely everything, but has the strength not to.

Her mosquito swarm probably came closer than anything else to actually hurting Saitama. He is amazingly resilient, but not completely invulnerable. He can bleed, so if the mosquitos could pierce his skin (granted, that's a pretty big if) they could drain him dry. Of course, he could have gotten rid of them in any number of ways, but Saitama prefers to solve problems by talking it out and/or punching.

Saitama would beat Goku, Supes, anyone at all because he is the ultimate gag character. He is the strongest. He wins by default, because that is his function. It's like asking if Arale or Squirrel Girl could beat a given character. It's not about whom Saitama could beat, it's about if he would hang out with them.
