MaRo responded that Emrakul was female. To be fair, MaRo said Nahiri and Emrakul, not just Emrakul. (Emrakul being human enough to have a gender identity at all is an awkward retcon, though.)

Thinking less about individual cards (don't know that I'd actually bother trying to do something as insane as that dude's Star Wars set) and more about mechanics that would: One Piece is a bit of a weird choice for this sort of thing because the major characters are so powerful; Luffy, even in East Blue, shouldn't really be threatened by any number of 1/1 soldier tokens in flavor terms. Though MtG has never been super-consistent about the flavor of power/toughness anyway (e.g., an elephant is equal to 3 squirrels).

They're meant to be game pieces, ones that care about really weird things and in a way that wouldn't work under the comp rules proper, but still game pieces. The focus on good gameplay is important, obviously, but I would have liked to see a bit more self-parody, instead of just the general humor they use in normal flavor text anyway. I guess there's a bit of that with the factions essentially being parodies of the Ravnica guilds, but even then, the Izzet, Rakdos, Gruul, and Azorius are often portrayed with a comic tone anyway. Though to be fair, most of the cards haven't been revealed yet.
