I don't believe the Rune of Earth is strictly necessary. The rune saves you a few turns of movement - probably 5-6, it looks like - and eliminates much of the logistical work. 2 of the U/W thrones are already within 4 provinces of the rune, and the other 2 are each within 4 tiles of an extant laboratory (or you could just build them).
