Mr. Nice! posted:didn’t a study just drop today that moderna is still like 80% effective for breakthroughs against delta and most breakthroughs are pfizer people? no it found pfizer about 76% effective. both vaccines are more or less identical, it's just that moderna has a slightly larger dose of the same juice but your takeaway from the moderna data should be that the vaccines are waning and nothing is safe right now when transmission is high. not a loving restaurant meal, not a "we'll just have the parents and family over for dinner (safely)", etc. vaccinated people are spreading. vaccinated people are getting sick. vaccinated people are not as protected as they think and they act they are. past about six months consider yourself totally unvaccinated again and act appropriately

I could swear there was an article in which Fauci uses the "There is no evidence masks work" phrasing in implicitly warning against masks in order to protect the mask supply, at the start of pandemic, and I'm sure it's out there, but I can't find it, and it's driving me a little nuts today. Can anyone else find it? All I could find regarding from January-March was https://www.usatoday.com/story/news...nds/4787209002/ https://www.factcheck.org/2021/02/scicheck-video-wrong-about-fauci-covid-19/ (scroll down to Fauci's comments on masks) The second link contradicts the first one, presumably because they missed that Fauci had already started to switch rhetoric by then.

So Moderna is a better choice if you care about "value" when being a vaccine junkie.

What I want to see is death counts. The long game for COVID isn't acute antibodies, it's gonna be memory cell mediated immunity. And if memory cell mediated immunity is decently strong than we should see reduced death counts in the face of increased case counts. Last year we were seeing 3000+ deaths per day around the same time we were seeing 200k cases per day, so that's kind of the crossover point I'm looking for.

Thanks to whoever suggested getting in touch with industrial supply. I think something's up with their order processing or w/e, the guy I talked to said it definitely should have been shipped out by now having ordered Friday. Definitely worth contacting them if you've placed an order and it hasn't gone out within a day or so.

Notorious R.I.M. posted:I don't think SARS-CoV-2 is evolutionarily "boxed in". On the Spike protein there are primarily neutralizing epitopes on the N-terminal domain and the Receptor binding domain. There are numerous immune-escpae mutations that show up on both of these (see: Recurrent Deletion Regions on the N-Terminal Domain and RBD escape mutations across three classes of monoclonal antibodies). So overall we're looking at several groups of mutations on the RBD that need to occur to escape several groups of antibodies, and several deletion regions on the N-Terminal Domain that escape several groups of antibodies. Adding one or two of these at a time will cause some antibody efficacy decrease, but adding all of them together is almost complete immune escape. Thanks for breaking this down so clearly. However regarding the spike-protein antibody escape mutations, you're pointing out that COVID can readily mutate it's spike to avoid antibodies but that was the exact subject of the video I linked. If I understand the video's content, which is dubious, they specifically discuss a set of 20 mutations that added together lead to total immune escape of immature, but not affinity matured, convalescent+vaccinated plasma. That was the original motivation for my saying COVID might be somewhat "boxed" in, there appear to be at least some antibodies it might not be able to fully escape according to these neutralization experiments. It's quite possible that was an unfounded interpretation, but what you've written here doesn't seem to directly contradict what I wrote. edit: Here's a preprint for the research described in the video: quote:High genetic barrier to escape from human polyclonal SARS-CoV-2 neutralizing antibodies Nocturtle has issued a correction as of 20:08 on Aug 11, 2021 |

And if memory cell mediated immunity is decently strong than we should see reduced death counts in the face of increased case counts. it's slightly an artifact of us not accurately tracking breakthrough cases, but uhhhh the CFR is not changing in vaccinated vs. unvaccinated groups. keep looking at recent state data, like massachusets (1.25% CFR of their 8000 breakthrough cases), LA, seattle, etc. it's all showing even in breakthrough cases we see 5-7% hospitalized and 1-1.5% die. that's exactly the same as OG covid against the unvaccinated population HOPEFULLY it's just because we have way, way more asymptomatic breakthrough cases than can be tracked... but who knows. it really could end up that the vaccines gave us a short window of immunity and now people have blown it all by acting immune and getting sick again

It's quite possible that was an unfounded interpretation, but what you've written here doesn't seem to directly contradict what I wrote. You can generate antibodies against more than one part of a virus, so as long as some of your antibodies can slow the virus down your body can adapt to the newly presented antigens and clear them out. It's why we can get sick with the cold, and also why it doesn't kill us. It takes a while to spin up a response from memory cells. The problem is when a novel pathogen like COVID shows up and kills 20 million people as part of that process. It's also a problem when you have a half-assed vaccination program putting enhanced evolutionary pressure on the virus to rapidly mutate away from the known antigens, and a half assed mitigation program that allows novel variants that might die in vivo to spread. Edit: It's also why a vaccine that only targets one specific antigen on the viral capsid may not be effective in the long-term when combined with the aforementioned half-assed programs that basically FORCE the virus to evolve, and render the vaccine and resulting memory cells with less or even nil efficacy.
